PT-141 (Bremelanotide) is a melanocortin receptor agonist peptide that enhances sexual arousal and libido by acting on the brain’s arousal centers.

Main Information

PT-141, also known as Bremelanotide, is a synthetic peptide that activates melanocortin receptors in the brain, particularly MC-3 and MC-4. These receptors play a central role in regulating sexual behavior, arousal, and libido.

Unlike ED drugs (like Viagra or Cialis), which work peripherally by increasing blood flow, PT-141 works centrally, directly stimulating the hypothalamus to enhance sexual desire and arousal. It is effective in both sexes and is FDA-approved (as a nasal spray) for female sexual interest/arousal disorder (FSIAD).

Its unique mechanism makes it suitable for individuals with:

  • Low libido (psychological or physiological)
  • Difficulty with arousal despite normal hormone levels
  • Stress-related sexual dysfunction
  • ED not responsive to PDE5 inhibitors

Scientific Background

PT-141, also known as Bremelanotide, is a synthetic peptide developed from the melanocortin pathway, specifically designed to enhance sexual desire and arousal by activating central melanocortin receptors (MC3R and MC4R) in the brain. Unlike traditional treatments for erectile dysfunction or libido loss (which act on blood flow), PT-141 works neurologically, targeting the hypothalamus to stimulate sexual motivation directly.

PT-141 is effective in both men and women, with benefits including:

  • Increased libido and arousal
  • Enhanced erectile function (in men)
  • Improved satisfaction and sensitivity
  • Useful in psychological or stress-related dysfunction, and in cases where PDE-5 inhibitors are ineffective

Originally observed during research on Melanotan II, PT-141 was developed as a standalone compound free from the tanning effects. It was FDA-approved in 2019 for hypoactive sexual desire disorder (HSDD) in premenopausal women and is widely used off-label in performance enhancement contexts for both sexes.

Usage Instructions

PT-141 is available in injectable and nasal spray formats. Timing is essential for optimal effect.

Injectable:

  • Reconstitute vial with 1–2 ml bacteriostatic water
  • Inject subcutaneously (abdomen or thigh)
  • Store reconstituted solution at 2–8 °C, use within 7–10 days

Nasal Spray:

  • Administer 1 spray per nostril (total 2 sprays = 1 dose)
  • Effects begin within 30–60 minutes, lasting up to 6–8 hours
  • Store refrigerated when not in use

Dosage Information

Men (Libido & Erectile Response):

  • Dose: 1–2 mg SC injection or full nasal dose
  • Use 45–60 min before sexual activity
  • Do not exceed 1 dose per 24 hours, or more than 8 doses/month

Women (Low Sexual Desire / FSIAD):

  • Dose: 1.75 mg nasal spray (FDA standard)
  • Use pre-intimacy, best results after several uses
  • Can be used 2–3× per week, depending on individual response

Stacking Potential:

  • May be combined with Oxytocin, Kisspeptin, or Testosterone microdosing
  • Often used in libido stacks or couples protocols

Important Warnings

  • Do not exceed recommended dosage. Overuse may increase the risk of side effects such as nausea or blood pressure elevation.
  • Not recommended for individuals with uncontrolled hypertension, cardiovascular disease, or a history of stroke, as PT-141 may cause transient increases in blood pressure and heart rate.
  • Common side effects may include nausea, flushing, headache, or injection site reactions. These are usually mild and self-limiting.
  • Avoid alcohol and CNS depressants when using PT-141, as they may interfere with arousal or increase side effects.
  • Use with caution if you are taking dopamine agonists, MAO inhibitors, or psychotropic medications, as PT-141 may influence dopaminergic and serotonergic systems.
  • Not approved for use during pregnancy or breastfeeding due to a lack of safety data.
  • PT-141 is for subcutaneous injection only. Avoid intravenous use.
  • Always store the peptide according to label instructions—typically refrigerated (2–8°C) after reconstitution.

Discontinue use and consult a healthcare provider if you experience chest pain, severe dizziness, or signs of allergic reaction.
